اثر عصاره هیدروالکلی آویشن زوفایی بر بلوغ آزمایشگاهی اووسیت گوسفند

چکیده

    بلوغ اووسیت و متعاقباً لقاح آن در آزمایشگاه از نقطه نظر تولید رویان حائز اهمیت زیادی است.  با توجه به این که آنتی­اکسیدان­ها تخریب کننده­ی رادیکال­های آزاد هستند، می­توانند بلوغ آزمایشگاهی اووسیت و کیفیت جنین را بهبود دهند.  بنابراین، هدف از مطالعه­ی حاضر ارزیابی اثرات عصاره­ی هیدروالکلی گیاه آویشن زوفایی روی بلوغ اووسیت گوسفند در آزمایشگاه است.  مجموعه­ی اووسیت- کومولوس استحصال شده از تخمدان میش به مدت 24 ساعت در محیط بلوغ آزمایشگاهی HTCM غنی شده با FSH، LH ، FBS، سیستئامین، پیرووات سدیم و  آنتی­بیوتیک (گروه کنترل) و در محیط بلوغ آزمایشگاهی فوق بدون سیستئامین ولی غنی شده یا غلظت­های مختلف عصاره هیدروالکلی آویشن زوفایی (µg/ml  1: گروه 1، µg/ml  10: گروه 2، µg/ml  50: گروه 3) کشت داده شدند.  بلوغ آزمایشگاهی و آغاز مجدد میوز در تمام گروه­های مورد آزمایش با تعیین میزان گسترش سلول­های کومولوس و تعداد اووسیت­های در مرحله متافاز تقسیم میوز II  بررسی شدند.  نتایج حاصل نشان داد که اووسیت­های تیمار شده با غلظت­های 10 و 50 از نظر میزان گسترش کل سلول­های کومولوس مشابه گروه کنترل بودند در حالی که گروه تیمار شده با غلظت 1 از این نظر کمتر از گروه کنترل بودند.  از نظر بلوغ هسته، گروه تیمار شده با غلظت 50 مشابه گروه کنترل بود در حالی که گروه­های تیمار شده با غلظت­های 1 و10 به طور معنی­دار از گروه کنترل کمتر بودند.  نتایج به دست آمده نشان داد که عصاره­ی آویشن زوفایی به صورت وابسته به مقدار، اثر مثبتی روی بلوغ اووسیت گوسفند دارد.  از این رو با افزایش غلظت عصاره میزان بلوغ اووسیت افزایش یافت که می­تواند به سبب ترکیبات آنتی­اکسیدان موجود در آن باشد.
