بررسی بیان ژن آنزیم‌های آنتی‌اکسیدانی در جوجه‌های گوشتی تغذیه شده با ویتامینE ، عصاره‌ی گیاه خرفه و پسته‌ی وحشی تحت استرس گرمایی

چکیده

    این تحقیق به منظور ارزیابی اثر عصاره­ی گیاهان خرفه، پسته­ی وحشی و ویتامین E بر بیان ژن‌های آنزیم‌های آنتی‌اکسیدانی (سوپراکسید دیسموتاز و کاتالاز) در جوجه‌های گوشتی تحت استرس گرمایی اجرا گردید.  بدین منظور از ۲۰۰ قطعه جوجه­ی گوشتی سویه­ی تجاری راس 308 در قالب طرح کاملاً تصافی با 5 تیمار، 4 تکرار و 10 قطعه جوجه در هر تکرار استفاده شد.  پنج تیمار آزمایشی عبارت بودند از:  1- جیره­ی شاهد (جیره­ی پایه بدون هیچ‌گونه افزودنی)، 2- جیره­ی پایه به همراه 200 میلی­گرم بر کیلوگرم ویتامین E، 3- جیره­ی پایه به همراه 100 میلی­گرم بر کیلوگرم عصاره­ی پسته­ی وحشی، 4- جیره­ی پایه به همراه 100 میلی­گرم بر کیلوگرم عصاره­ی خرفه، 5- جیره­ی پایه به همراه 100 میلی­گرم بر کیلوگرم عصاره­ی پسته­ی وحشی و 100 میلی­گرم بر کیلوگرم عصاره­ی خرفه بود.  در انتهای آزمایش دو مرغ از هر تکرار کشتار شده و کبد آن­ها سریعاً جدا گردید و به همراه ازت مایع به آزمایشگاه منتقل گردید.  بیان ژن‌های آنزیم‌های سوپر اکسید دیسموتاز و کاتالاز به روش Real-time qPCR ارزیابی شد.  در این روش ژن بتااکتین به عنوان ژن خانگی (منبع) جهت نرمال نمودن داده‌ها استفاده شده است.  نتایج نشان دهنده­ی این است که بیان ژن‌های آنزیم‌های سوپر اکسید دیسموتاز و کاتالاز بین تیمارها اختلاف معنی‌داری دارند.  همچنین در بیان ژن آنزیم‌های آنتی‌اکسیدانی سوپراکسید دیسموتاز و کاتالاز، بیش­ترین میزان را تیمار 5 داشت.  البته بیان این ژن‌ها در سایر تیمارها (تیمار ۲ و ۳ و ۴) نسبت به کنترل افزایش معنی‌داری را نشان داد.  بنابراین نتایج نشان داد که ترکیب عصاره‌ی پسته‌ی وحشی و گیاه خرفه در کنار هم می‌تواند تأثیر به سزایی بر بیان ژن‌های آنزیم‌های آنتی‌اکسیدانی در شرایط استرس گرمایی داشته باشد.
