شناسائی ملکولی گونه های بیماریزای آناپلاسما در گاوهای استان مازندران

چکیده

    هدف از این مطالعه تعیین گونه‌های آناپلاسما در گاوهای استان مازندران بود.  برای این منظور تعداد 105 نمونه خون از طریق رگ وداج گاو به طور تصادفی از نقاط مختلف استان مازندران جمع‌آوری گردید.  در ابتدا DNA استخراجی از نمونه‌های خونی با جفت آغازگری که قطعه­ی 1468 جفت بازی از ژن 16S rRNA جنس آناپلاسما را تکثیر می‌کرد، تکثیر شد.  29 نمونه (6/27 درصد) از 105 نمونه گاوی در اولین PCR و nested-PCR از نظر جنس آناپلاسما مثبت شدند.  تمامی نمونه‌های مثبت با nested-PCR اختصاصی از نظر وجود آناپلاسما فاگوسیتوفیلم، آناپلاسما بوویس و آناپلاسما سنتراله (سویه آموری) بررسی شدند و 21 نمونه (20 درصد) از نظر آناپلاسما فاگوسیتوفیلم، 12 نمونه (4/11 درصد) از نظر آناپلاسما بوویس و 1 نمونه (1 درصد) از نظر آناپلاسما سنتراله (سویه آموری) مثبت تشخیص داده شد.  DNA استخراجی نمونه‌های مثبت با استفاده از جفت آغازگرهای اختصاصی که قطعه 866 جفت بازی از ژن msp4 آناپلاسما مارژیناله را تکثیر می‌کرد، تکثیر شد که 5 نمونه (8/4 درصد) از 105 نمونه گاوی از نظر آناپلاسما مارجیناله مثبت بودند.  این مطالعه اولین تشخیص ملکولی گونه‌های آناپلاسما در گاوهای استان مازندران بوده است.  نتایج نشان می‌دهد که در بین درصد آلودگی گاو به آناپلاسما فاگوسیتوفیلم، در فصول مختلف سال و نوع دامداری، اختلاف معنی‌دار وجود دارد.  همچنین از نظر درصد آلودگی به آناپلاسما بوویس، از میان متغیر‌های مورد بررسی، بین فصول مختلف سال و نوع دامداری اختلاف معنی‌دار وجود دارد.
