مقایسه رقیق کننده حاوی پلاسمای زرده تخم کبوتر، مرغ و ترکیب آن‌ها جهت نگهداری منی قوچ در دمای 4 درجه سانتی‌گراد

چکیده
    مطالعه حاضر به منظور بررسی اثر پلاسمای زرده تخم کبوتر و مرغ در مقایسه با ترکیب پلاسمای زرده کبوتر+ مرغ در رقیق کننده تریس-اسید سیتریک-فروکتوز جهت حفظ کیفیت منی قوچ به صورت مایع-سرد انجام شد.  نمونه­های منی با استفاده از واژن مصنوعی از چهار قوچ نژاد قزل دوبار در هفته جمع­آوری شد و در صورتی که معیارهای مورد نظر را داشت با هم مخلوط و در مطالعه حاضر مورد استفاده قرار گرفت.  رقیق کننده تریس-سیتریک اسید- فروکتوز با استفاده از پلاسمای زرده تخم کبوتر (28 درصد، حجمی/حجمی)، تخم مرغ (28 درصد) و ترکیب پلاسمای زرده تخم کبوتر (7، 14 و 21 درصد) و مرغ (21، 14 و 7 درصد) تهیه و جهت آزمایش مورد استفاده قرار گرفت.  متعاقب رقیق­سازی نمونه­ها، شاخص­های حرکت کلی و پیشرونده رو به جلو اسپرم (توسط سیستم آنالیز کامپیوتری اسپرم)، زنده­مانی و انسجام غشای اسپرم در ساعات صفر، 24، 48 و 72 مطالعه بعد از سرد کردن (4 درجه سانتی­گراد)، ارزیابی شد.  همچنین، مقدار مالون­دی­آلدهید، به عنوان شاخص اکسیدانتی در ساعت­های مذکور اندازه­گیری شد.  نتایج آزمایش نشان داد که حرکت پیشرونده رو به جلو (در ساعات 24، 48 و 72) و زنده­مانی (در ساعت 72) در رقیق کننده حاوی 28 درصد پلاسمای زرده تخم کبوتر به طور معنی­دار بالاتر از گروه­های دریافت کننده ترکیب پلاسمای زرده تخم کبوتر+مرغ بود.  همچنین انسجام غشای پلاسمایی اسپرم در گروه دریافت کننده 28 درصد پلاسمای زرده تخم کبوتر در ساعات 24، 48 و 72 بالاتر از گروه­های ترکیبی با غلظت 14 و 21 درصد پلاسمای تخم مرغ بود.  غلظت مالون­دی­آلدهید بین گروه­های مطالعه اختلاف معنی­دار نشان نداد.  در نتیجه ترکیب پلاسمای تخم مرغ و کبوتر به انداره رقیق کننده حاوی پلاسمای زرده تخم کبوتر و پلاسمای زرده تخم مرغ (هر کدام به تنهایی) جهت نگهداری منی قوچ به صورت مابع-سرد کارا نمی­باشد.
