تعیین هویت مولکولی و آنالیز فیلوژنتیکی ویروس طاعون نشخوارکنندگان کوچک در سال‌های 95 و 97 در استان قم

چکیده

    طاعون نشخوارکنندگان کوچک (PPR) بیماری ویروسی بسیار واگیر، شدید و کشنده نشخوارکنندگان کوچک است.  چهار دودمان (Lineage) ویروس PPR بر اساس تحلیل توالی ژن نوکلئوپروتئین (N) و فیوژن (F) شناخته شده است.  هدف از این مطالعه، بررسی تعیین هویت مولکولی و آنالیز فیلوژنتیکی ویروس طاعون نشخوارکنندگان کوچک در رخدادهای PPR سال­های 95 و 97 در­ استان قم بوده است.  10 نمونه­ی بافتی (غدد لنفاوی مزانتر) پس از کالبدگشایی از حیوانات مشکوک بالینی جمع­آوری شد و وجود ویروس PPR  با روش RT-PCR یک مرحله­ای مورد بررسی قرار گرفت.  بررسی توالی قطعه­ای از  ژن نوکلئوپروتئین (N) و آنالیز فیلوژنتیکی نمونه­هایی که در RT-PCR مثبت بودند جهت تعیین دودمان جدایه­ها انجام گرفت.  از 10 نمونه بالینی مورد بررسی، 40 درصد آن­ها بر اساس بررسی اسید نوکلئیک ویروس با روش RT-PCR مثبت بودند.  آنالیز توالی نوکلئوتیدی و تجزیه فیلوژنتیکی جدایه­ها نشان داد که جدایه­های به دست آمده در رده­ی آسیایی ­(دودمان ­IV) ویروس PPR قرار دارد.  نتایج فیلوژنتیک این مطالعه، ­حضور ویروس PPR دودمان ­IV را در میان دام­های مبتلا به طاعون نشخوارکنندگان کوچک استان قم، تأیید می‌کند.  بنابراین این مطالعه نشان می­دهد که جدایه‌های PPRV ایران به دودمان چهارم مربوط می­شوند و با جدایه­های PRPV پاکستان، تاجیکستان و هند ارتباط نزدیک دارند.  توصیه می­شود به منظور ترسیم دقیق­تری از وضعیت حضور و چرخش دودمان یا دودمان‌های ویروس PPR در ایران، مطالعات بیش­تری از دیگر مناطق کشور به خصوص نواحی مرزی انجام گیرد.
