Achieving the results of total microbial count in minimum time is really important for confidence from the hygienic quality of products. Therefore, impedance splitting method as a new technique for this purpose was considered in order to receiving the results in less time and as soon as possible. The main purpose of this study, was to evaluate the correlation between impedance detection time (IDT in hrs), total microbial population (log10 N) and titrable acidity (0D) of raw and pasteurized milk. During 6 months, 100 samples (50 samples of raw and 50 samples of pasteurized milk) were collected from different areas of Ahvaz and examined under sterile conditions. The total microbial count by pour plate technique and impedance splitting method, also measuring milk titrable acidity were carried out based on the recommendations of Iran's Standard Institute and Industrial Investigation. Then, the calibration curves of 3 methods and their equations were obtained by using Excel software.  The calibration curves of methods were elaborated for total microbial count and impedance detection time, demonstrating a good correlation between the two methods (95.29% for raw and 95.09% for pasteurized milk samples). According to the calibration curves, the correlation between impedance detection time and  titrable acidity was 88.83% for raw and 85.4% for pasteurized milk samples, also the correlation between total microbial count and titrable acidity was 94.14% for raw and 93.27% for pasteurized milk samples. Therefore, impedance measurement which is a more rapid, automated and less laborious method than conventional total microbial count technique could be used like some developed countries as an alternative method for the rapid measuring the total microbial loads in foods instead of conventional methods.  }, keywords = {Milk,Impedance,Microbial load,acidity}, title_fa = {بررسی روش اندازه‌گیری بار میکروبی شیر خام و پاستوریزه با استفاده از اندازه‌گیری مقاومت الکتریکی (امپدانس) و تطابق آن با اسیدیته قابل تیتر شیر}, abstract_fa = {    شمارش کلی بار میکروبی نمونه‌های شیر به روش مرجع پورپلیت و مطابقت نتایج حاصله با استانداردهای موجود، از جمله کارهای آزمایشگاهی معمول در تمامی کارخانجات صنایع شیر است.  از سوی دیگر، سرعت دستیابی به نتایج در اسرع وقت از جمله نکات ویژه و مدنظر کارخانجات به منظور اطمینان از کیفیت محصول تولیدی و در حال توزیع می‌باشد.  لذا با تکیه بر این موضوع، بهره‌گیری از تکنیک امپدانس در ارزیابی بار میکروبی شیرهای خام و پاستوریزه مد نظر واقع شد که با صرف وقت کمتر، دستیابی سریع‌تر به نتایج را میسر می‌سازد.  همچنین در این مطالعه، مطابقت نتایج حاصله از روش امپدانس با نتایج به دست آمده از روش مرجع پورپلیت و نیز اسیدیته قابل تیتر در شیر مورد بررسی واقع گردید.  در طی این بررسی، تعداد 100 نمونه شیر (50 نمونه پاستوریزه و 50 نمونه خام) تهیه شد و با استفاده از روش امپدانس و نیز روش مرجع پورپلیت مورد ارزیابی از نظر شمارش کلی بار میکروبی قرار گرفتند.  همچنین مقدار اسیدیته شیر بر اساس درجه دورنیک اندازه‌گیری شد.  روش پورپلیت، اسیدیته شیر و روش امپدانس، بر اساس دستورالعمل‌های موسسه استاندارد و تحقیقات صنعتی ایران انجام پذیرفت و سپس با استفاده از نتایج، منحنی‌های مربوط به انطباق سه روش با یکدیگر و معادله منحنی‌های مذکور با استفاده از روش‌های آماری و نرم‌افزار Excel به دست آمد.  بر اساس نتایج حاصل از این تحقیق، میزان تطابق روش امپدانس با روش مرجع پورپلیت در شیرهای پاستوریزه 01/95% و در شیرهای خام 29/95% بود.  همچنین میزان تطابق روش امپدانس با مقدار اسیدیته قابل تیتر در شیرهای خام براساس درجه دورنیک 83/88% و در شیرهای پاستوریزه 4/85% و نیز میزان تطابق مقدار بار باکتریایی شیر با مقدار اسیدیته در شیرهای خام و پاستوریزه به ترتیب 14/94% و 27/93% به دست آمد.  بنابر نتایج مطالعه حاضر، تکنیک امپدانس به عنوان جایگزینی برای روش‌های مرسوم قدیمی می تواند مورد استفاده باشد.}, keywords_fa = {شیر,امپدانس,بار میکروبی,اسیدیته}, url = {https://www.ivj.ir/article_2981.html}, eprint = {https://www.ivj.ir/article_2981_c93ddc0d054b78c347d1a79f80c3fdc4.pdf} }
