A total of 253 blood samples were collected via the jugular vein from healthy cattle, randomly. The extracted DNA from blood cells was amplified by Babesia-all primers, which amplify an approximately 400bp DNA fragment from the region of the 18S rRNA gene from various members of the genus Babesia. All cattle positive samples were further analysed for the presence of B. bigemina and B. bovis by specific semi-nested PCR. B. bigemina and B. bovis were identified by specific semi-nested PCR in 0% and 65.2% of cattle blood samples, respectively. Chi-square tests were used to compare molecular prevalence values relative to Season, Farm, Type, Hygiene, Vectors, Use a disposable needle, Age, and Milk Yield. Among these factors, seasons and vectors were found to have significantly different in the prevalence. The significant major risk factors of B. bovis in cattle were identified as season, hygiene, and vectors by the univariate analysis. Moreover, multivariable logistic regression analysis revealed a statistically significant association of the prevalence of B. bovis with the season. The examination of 50 microscopic fields showed 59.39% sensitivity and 100% specificity compared to molecular examination. The Kappa coefficient between molecular and microscopy (50 fields) techniques indicated a moderate level of agreement (Kappa= 0.504). This study is the first molecular detection of Babesia species from cattle in the south of Isfahan Province, Iran. Further researches are needed to determine the vectors, vector-host interactions and genotypic variants that may affect the presence and distribution of Babesia species in Iran.}, keywords = {Iran,Isfahan Province,Babesia species,Cattle,Molecular Detection}, title_fa = {تشخیص ملکولی و شیوع بابزیا بوویس در گاوهای شهرستان شهرضا، جنوب استان اصفهان}, abstract_fa = {    هدف از این مطالعه تعیین گونه‌های بابزیا در گاوهای شهرستان شهرضا، در جنوب استان اصفهان بود.  در مجموع تعداد 253 نمونه خون از طریق ورید وداج گاوهای به ظاهر سالم به طور تصادفی اخذ شد.  در ابتدا DNA استخراجی از نمونه‌های خونی با جفت آغازگری که قطعه حدود 400 جفت بازی از ژن 18S rRNA جنس بابزیا را تکثیر می‌کرد، تکثیر شد.  تمامی نمونه‌های مثبت گاوی با nested-PCR semi اختصاصی از نظر وجود بابزیا بایجمینا و بابزیا بوویس بررسی شدند و آلودگی نمونه‌های گاوی از نظر بابزیا بایجمینا و بابزیا بوویس به ترتیب صفر و 2/65 درصد تشخیص داده شد.  آزمون مربع کای جهت مقایسه میزان شیوع نسبت به فصل سال، نوع دامداری، بهداشت دامداری، ناقلین بندپا، استفاده از سرسوزن مجزا، سن دام و تولید شیر انجام شد.  در مقایسه فراوانی گونه بابزیا بوویس در گاو بین دو فصل نمونه‌گیری شده و حضور ناقلین اخلاف معنی‌داری مشاهده شد.  در آنالیز لجستیک تک متغیره فصل، بهداشت دامداری و ناقلین بندپا به عنوان عوامل خطر مهم برای ابتلا به بابزیا بوویس مشخص شدند.  در مدل لجستیک چند متغیره تنها فصل به عنوان مهم‌ترین عامل در ابتلا به بابزیا بوویس معرفی شد.  در مقایسه با آزمون ملکولی، حساسیت و ویژگی آزمون میکروسکوپی به ترتیب 39/59 و 100 درصد تعیین گردید.  محاسبه ضریب کاپا بین آزمون ملکولی و میکروسکوپی بابزیا (50 فیلد) نشان‌دهنده سطح متوسط توافق دو آزمون بود (Kappa= 0.504).  این مطالعه اولین مطالعه تشخیص ملکولی گونه‌های بابزیا در گاوهای منطقه جنوب اصفهان است.  تحقیقات دیگری در جهت شناسایی ناقلین، اثر متقابل میزبان- ناقل و شناسایی واریته­های ژنتیکی که ممکن است حضور و گسترش گونه‌های بابزیا را در گاوهای ایران تحت تأثیر قرار دهند مورد نیاز است.}, keywords_fa = {استان اصفهان,ایران,گونه‌های بابزیا,گاو,تشخیص ملکولی}, url = {https://www.ivj.ir/article_148235.html}, eprint = {https://www.ivj.ir/article_148235_ffec82b38b936f14fa49ee8b00a62b74.pdf} }
