Concerns remain in place as the elusive MAP epidemiology and its potential zoonotic nature continue to challenge veterinarians across the world. Almost half a century since its initial reports, Ptb is still not a notifiable disease in Iran leading to the unfortunate lack of a public-funded control scheme. We undertook this study to extend our knowledge of the MAP population genetic in diverse hosts in Iran. During seven months, 35 MAP isolates collected from milk and feces obtained from sheep, goat and cattle originating 8 provinces plus a further eleven archived MAP isolates were genetically interrogated. The Collins strategy was conducted on examined isolates to differentiate between cattle and sheep types of MAP bacteria. The study was further expanded by RFLP-IS900 analysis using PstI in search for genetic diversity. The findings of Collins strategy showed that all animal species covered by the study were exclusively infected by 'cattle' strains. In genotyping, on the other hand, RFLP-IS900 analysis yielded 10 largely similar genotypes with five of them infecting multiple hosts. Nevertheless, no trace of varying profiles was detected in individual farms and conspecifics. These data, taken together with earlier observations by others, raise the question of the mysterious absence of sheep-type strains in Iran. Circulation of highly similar strains in Iranian farm ruminants might be an indication of a homogenic MAP population with a tendency for diversification started only recently. The extent of temporal and spatial characteristics of such diversification is still unclear and left for future works.}, keywords = {Paratuberculosis,Mycobacterium avium,Sheep type strain}, title_fa = {پروفایل ژنتیکی جدایه های مایکوباکتریوم ایویوم زیرگونه پاراتوبرکولوزیس جمع آوری شده از مدفوع و شیر گاو، گوسفند و بز در ایران، فقدان خارج از انتظار سویه‌های تیپ گوسفندی پاراتوبرکولوزیس در ایران}, abstract_fa = {    مایکوباکتریوم ایویوم زیرگونه­ی پاراتوبرکولوزیس (MAP) عامل ایجاد پاراتوبرکولوزیس (Ptb) در جهان شناخته می­شود.  پاراتوبرکولوزیس یک بیماری مزمن بدون درمان در دستگاه گوارش نشخوارکنندگان می­باشد. اپیدمیولوژی پیچیده MAP همراه با پتانسیل آن در ایجاد بیماری در انسان همچنان به عنوان یک چالش جهانی دامپزشکان محسوب می­گردد.  تقریباً نیم قرن پس از شناسایی اولین موارد این بیماری، هنوز گزارش وقوع پاراتوبرکولوزیس در ایران الزامی نمی­باشد و این امر زمینه­ی فقدان یک برنامه­ی جامع کشوری کنترل بیماری را فراهم نموده است.  مطالعه­ی حاضر به منظور افزایش دانش موجود از ژنتیک جمعیت MAP در میزبان­های مختلف در ایران انجام پذیرفته است.  در مدت هفت ماه تعداد ۳۵ جدایه از شیر و مدفوع گوسفند، بز و گاو در ۸ استان کشور همراه با ۱۱ جدایه آرشیو شده MAP بررسی گردیدند.  استراتژی کالینز برای تشخیص افتراقی میان تیپ­های گاوی این باکتری­ها استفاده شد.  مطالعه با بکارگیری RFLP-IS900 با استفاده از آنزیم PstI گسترده­تر گردید.  استراتژی کالینز نشان داد همه­ی دام­های تحت بررسی فقط با سویه­های تیپ گاوی آلوده شده بودند.  در تایپینگ ژنتیکی یافته­های RFLP-IS900نشان دهنده­ی وجود ۱۰ تیپ با شباهت زیاد بود که در ۵ مورد میزبان­های مختلف را آلوده نموده بودند.  در عین حال نشانه­ای از وجود بیش از یک سویه در یک فارم و یا در یک میزبان مشاهده نگردید.  یافته­های این مطالعه در کنار مشاهدات گزارش شده توسط دیگران این سوال را بر می­انگیزند که چرا در ایران سویه­های تیپ گوسفندی مشاهده نمی­شوند.  گردش سویه­های بسیار مشابه MAP در میان نشخوارکنندگان اهلی ایران می­تواند نشانه­ای از وجود یک جمعیت یکنواخت ژنتیکی این پاتوژن باشد که اخیراً متحمل تحولات ایجاد کننده تنوع ژنتیکی شده است.  دامنه­ی مکانی و زمانی این گونه تغییرات در حال حاضر نامشخص و آگاهی از آن­ها نیازمند مطالعات آتی خواهد بود.}, keywords_fa = {پاراتوبرکولوزیس,مایکوباکتریوم ایویوم,سویه تیپ گوسفندی}, url = {https://www.ivj.ir/article_128897.html}, eprint = {https://www.ivj.ir/article_128897_be275b601a1fdefee425c9bbb856bbad.pdf} }
