The blood samples were collected from 41 multiparous (without pre and postpartum clinical diseases) Holstein cows in a modern dairy herd. DNA was extracted from whole blood and the β-casein genotype was detected by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis using BstDE I (Dde I) enzyme based on the replacement of the proline by histidine at position 67 β-casein protein in A1 milk. The PCR products sequenced for each genotype which used as control samples to validate the RFLP findings. The reproductive indices between cows with different genotypes were compared by Kruskal–Wallis test. The results of the present study indicated that A1A1, A1A2 and A2A2 genotype had a frequency of 13 (31.7%), 21 (51.2%) and 7 (17.1%). No significant difference was found in calving to first service interval, days open and service per conception indices between studied cows with different β-casein genotypes (P > 0.05). As results, it seems that the identification and proliferation of cows with the allelic form of A2 of β-casein gene can be considered without adverse effects on the fertility of dairy cows. However, further studies are needed to investigate the relationship between this genotype and milk yield of cows in the dairy herds.}, keywords = {Fertility,β-casein,Genotype,dairy cow}, title_fa = {ارتباط چندشکلی جایگاه ژنی بتاکازئین شیر با صفات بازدهی تولید مثلی در گاوهای شیری هلشتاین}, abstract_fa = {    هدف از انجام مطالعه­ی حاضر، شناسایی گاوهای شیری هلشتاین در دو گروه ژن­های کدکننده­ی پروتئین­های A1 و A2 بتاکازئین شیر با استفاده از روش RFLP PCR- و ارتباط آن­ها با شاخص­های تولیدمثلی بود.  بدین منظور از 41 رأس گاو هلشتاین چندشکم‌زا (فاقد هرگونه بیماری­های بالینی پیرامون زایمان)، از یک گاوداری صنعتی شیری در اطراف شهرستان شیراز نمونه خون گرفته شد.  پس از استخراج DNA کل از خون کامل، تکثیر ناحیه دارای چندشکلی (با جایگزینی اسید آمینه پرولین با هیستیدین در جایگاه 67 اسید آمینه پرتئین بتاکازئین در A1)، از هضم آنزیمی BstDE I (Dde I) جهت تعیین ژنوتیپ ژن بتا کازئین استفاده گردید.  جهت اطمینان از روش کار و هضم آنزیمی از محصولات PCR تعیین ترادف شده به عنوان کنترل استفاده شد.  مقایسه­ی شاخص­های تولیدمثلی (فاصله­ی زایمان تا اولین تلقیح، روزهای باز و تعداد تلقیح به ازای آبستنی) بین گاوهای با ژنوتیپ‌های مختلف با استفاده از آزمون کروسکال والیس انجام شد.  نتایج مطالعه­ی حاضر نشان داد که تعداد 13 (7/31 درصد)، 21 (2/51 درصد) و هفت (1/17 درصد) رأس از گاوها به ترتیب دارای ژنوتیپ A1A1، A1A2 و A2A2 بودند.  مقایسه میانگین فاصله­ی زایمان تا اولین تلقیح، روزهای باز و تعداد تلقیح به ازای آبستنی بیان­گر عدم تفاوت معنی­دار بین گاوهای مورد مطالعه دارای ژنوتیپ­های مختلف بتاکازئین بود.  بر اساس نتایج، چنین به نظر می­رسد که شناسایی و تکثیر گاوهای دارای ژن بتاکازئین A2 (A1A2 یا A2A2) بدون نگرانی از تأثیر منفی آن بر باروری گاوهای شیری، می‌تواند مدنظر قرار گیرد.  با این وجود جهت بررسی ارتباط این ژنوتیپ و میزان تولید شیر در گله­های گاو شیری نیاز به مطالعات گسترده‌تری در این زمینه وجود دارد.}, keywords_fa = {باروری,بتاکازئین,ژنوتیپ,گاو شیری}, url = {https://www.ivj.ir/article_125871.html}, eprint = {https://www.ivj.ir/article_125871_989ccce5eace99ad634bd92077f80754.pdf} }
