The purpose of this study was to determine sequences and single nucleotide promotor mutations and heat shock gene 70 and to investigate the effect of single nucleotide mutations on the three-dimensional structure and thermal shock protein function of Khuzestan native chicken strain using bioinformatics methods. To do this research, 20 native blood samples were taken from different locations of Khuzestan province. After DNA extraction, using several primers, promoter and heat shock protein 70 gene were amplified and sequenced. Three mutations were observed throughout the complete heat shock protein 70 gene. The first mutation in nucleotide position 259 (A259G) was observed, which guanine replaced adenine. The second mutation occurred at the nucleotide position of 277 (C277G), where guanine was replaced by cytosine. The third mutation occurred at the nucleotide position of 1749 (C1749G), where guanine was replaced by cytosine. In the first and second mutations, there is no change in the amino acid, but the mutation at position 1749 causes the serum amino acid to become tryptophan. The mutation occurred at the position of 583 amino acid heat shock protein but did not change the 3D building and its performance. This amino acid change causes more stability in the heat shock protein 70 and maybe one of the reasons for the resistance of the native to the heat.}, keywords = {sequencing,Single nucleotide mutation,Promotor,Heat shock protein 70,Native chicken}, title_fa = {تعیین توالی و جهش‌های تک نوکلئوتیدی پروموتور و ژن پروتئین شوک حرارتی ۷۰ مرغ‌های بومی خوزستان}, abstract_fa = {    پروتئین شوک حرارتی ۷۰ یک چاپرون است که در پاسخ به استرس بیان می‌شود.  هدف از این تحقیق تعیین توالی و جهش‌های تک نوکلئوتیدی پروموتور و ژن پروتئین شوک حرارتی ۷۰ و بررسی اثر جهش‌های تک نوکلئوتیدی بر ساختار سه بعدی و عملکرد پروتئین شوک حرارتی ۷۰ مرغ بومی استان خوزستان با استفاده از روش­های بیوانفورماتیکی بود.  جهت انجام این تحقیق از ۲۰ قطعه مرغ بومی از مناطق مختلف استان خوزستان خون­گیری انجام شد.  پس از استخراج DNA، با استفاده از چندین پرایمر، پروموتور و ژن پروتئین شوک حرارتی ۷۰ تکثیر گردیده و توالی‌یابی شد.  سه جهش در طول کامل ژن پروتئین شوک حرارتی مشاهده گردید.  اولین جهش در جایگاه نوکلئوتید ۲۵۹ (A259G) مشاهده شد که گوانین جایگزین آدنین شده بود.  جهش دوم در جایگاه نوکلئوتید ۲۷۷ (C277G) اتفاق افتاده بود که گوانین جایگزین سیتوزین شده بود.  جهش سوم در جایگاه نوکلئوتید ۱۷۴۹ (C1749G) اتفاق افتاده که گوانین جایگزین سیتوزین شده بود.  در جهش اول و دوم تغییری در اسیدآمینه ایجاد نمی‌شود اما جهش در جایگاه ۱۷۴۹ سبب تبدیل شدن اسیدامینه­ی سرین به تریپتوفان می‌شود.  این جهش در جایگاه ۵۸۳ اسید امینه پروتئین شوک حرارتی رخ داده اما تغییری در ساختمان سه بعدی و عملکردش ایجاد نمی‌کند.  این تغییر اسیدآمینه سبب پایداری بیش­تر در پروتئین شوک حرارتی ۷۰ شده و ممکن است یکی از علل مقاومت مرغان بومی به گرما باشد.}, keywords_fa = {تعیین توالی,جهش تک نوکلئوتیدی,پروموتور,پروتئین شوک حرارتی ۷۰,مرغ بومی}, url = {https://www.ivj.ir/article_101927.html}, eprint = {https://www.ivj.ir/article_101927_6400bb38f1657a2034ef5155df549e44.pdf} }
