مقایسه تکثیر و پاساژهای متوالی تاکی زوئیت های نئوسپورا کنینوم در سل لاین های Vero و TLI

چکیده

نئوسپورا کنینوم یک عامل بیماریزای مهم در گاو و سگ می باشد. امروزه به طور گسترده ای از کشت سلولی به منظور جداسازی و تکثیر نئوسپورا برای اهداف مختلفی استفاده می شود. در مطالعه حاضر، میزان یکسانی از تاکی زوئیت انگل در مجاورت میزان یکسانی از سل لاین های Vero (سلول اپیتلیال کلیه میمون سبز آفریقایی) و سل لاین TLI (لنفوسیت ترانسفورم شده با تیلریا) قرار گرفت و به مدت سه ماه پاساژ متوالی داده شدند. به منظور ارزیابی میزان حدت تاکی زوئیت های به دست آمده از هر سل لاین، دوزهای مختلف انگل (5000، 10000 و 15000 تاکی زوئیت در هر میکرولیتر)، به تخم مرغ جنین دار تزریق شد و میزان تلفات ثبت و از بافتهای مغز، قلب و کبد آنها جهت مطالعات هیستوپاتولوژی و مولکولی نمونه برداری انجام شد. نتایج این مطالعه نشان داد که سل لاین TLI توان تکثیر مناسبی را برای نئوسپورا نسبت به سل لاین Vero دارد و اختلاف آماری معنی داری در تعداد تاکی زوئیت های رها شده از دو سل لاین مشاهده نشد (p> 0.05). نتایج میزان تلفات، مرگ و میر 100% در تخم مرغ های تلقیح شده به وسیله دوز های مختلف تاکی زوئیت های به دست آمده از هر دو سل لاین را نشان داد که البته این میزان تلفات، وابسته به دوز تلقیح شده بود؛ بدین صورت که در دوزهای بالاتر، تلفات زودتر مشاهده شد. بررسی های هیستوپاتولوژیک، هپاتیت و میوکاردیت غیرچرکی تک هسته ای را نشان داد. با مطالعات مولکولی و ایمنوهیستوشیمی حضور آنتی ژن و ژنوم نئوسپورا در بافت های مختلف تائید شد. یافته های این پژوهش، سل لاین مناسبی را جهت تکثیر و پاساژ متوالی نئوسپورا کنینوم معرفی می کند که می تواند در دستیابی به روش سریعتر و موثرتر جهت تهیه واکسن زنده و همچنین صرفه جویی در زمان و هزینه ها بسیار کمک کننده باشد.
