تعیین دوزاژ بهینه بالینی فلورفنیکل خوراکی در ماهیان قزل آلای رنگین کمان دچار استرپتوکوکوزیس/ لاکتوکوکوزیس تجربی و بررسی باقیمانده دارویی در کبد و ماهیچه آنها

چکیده

بروز پدیده­ی مقاومت به آنتی­بیوتیک­ها در عوامل باکتریایی بیماری­زا در پی مصرف بی­رویه از یک سو و اهمیت به کار گیری آنتی­بیوتیک­ها در همه­گیری­های همراه با شکست­های سنگین اقتصادی از سویی دیگر، سبب شده است تا یافتن دوز بهینه­ی بالینی اهمیت خاصی داشته باشد.  پژوهش حاضر با هدف تعیین دوزاژ بهینه­ی بالینی آنتی­بیوتیک به روش خوراکی در ماهیان قزل­آلای رنگین­کمان مبتلا به بیماری استرپتوکوکوزیس/ لاکتوکوکوزیس و همچنین ردیابی باقیمانده­ی دارویی در کبد و ماهیچه­ی آن­ها انجام شد.  بدین منظور، نخست میانه­ی دوز کشنده (LD50) باکتری­های استرپتوکوکوس اینیایی و لاکتوکوکوس گارویه برای ماهیان به کمک ثبت تلفات روزانه و محاسبه رگرسیون پروبیت به دست آمد.  این دوزها برای آزمایش تعیین میانه­ی دوز مؤثر (ED50) در گروه­های کنترل و تحت درمان خوراکی با دوزاژهای mg/kg-1 BW 5، 10، 20 و 40 فلورفنیکل به مدت 10 روز متوالی به کار گرفته شدند.  1، 5 و 10 روز پس از دوره­ی درمانی، از کبد و ماهیچه­ی ماهیان نمونه­برداری شده و پس از مراحل آماده­سازی و تهیه­ی استانداردها، سنجش غلظت باقیمانده­ی دارویی از طریق کروماتوگرافی مایع با کارایی بالا (HPLC) انجام شد.  مقادیر ED50 فلورفنیکل خوراکی برای L. garvieae و S. iniae به ترتیب برابر با mg/kg BW 52/14 و 49/15 به مدت 10 روز متوالی به دست آمد.  بیشینه­ی حد باقیمانده (MRL) در روز دهم نمونه­برداری در کبد و ماهیچه به ترتیب µg/g BW (064/0±)461/0 و (103/0±)622/0 و به طور معنی­داری کم­تر از مقدار مجاز استاندارد آژانس اروپایی ارزیابی محصولات دارویی بود.  دوزاژ بهینه­ی بالینی فلورفنیکل در ماهیان مبتلا به این بیماری معادل میانگین میانه­ی دوز مؤثر دارو برابر با mg/kg BW 15 برای 10 روز متوالی شناخته شده است که پس از به کارگیری این دوزاژ حداقل 10 روز زمان لازم است تا مصرف کننده بتواند از این ماهیان استفاده خوراکی کند.
