Samples of present research collected from wetlands of Boushehr province in south of Iran and obtained from Razi Vaccine and Serum Research Institute, Shiraz, Iran. Virus isolation and characterization of the samples were performed at the Razi institute. The biological properties and pathogenicity of five NDV isolated were studied by intra cerebral pathogenicity index (ICPI), mean death time (MDT) value and reverse transcription-polymerase chain reaction (RT-PCR) methods. The PCR was used to amplify 362bp the cleavage site of all isolates. Sequencing of PCR products was performed to identify nucleotides and deduce amino acids. Alignment and phylogenetic analysis of F protein (cleavage site) was performed by analysis was done by software Megalign5, Blast and Bioedit. MDT and ICPI method showed that all of 5 samples were detected as lentogenic strain with low virulent and so, in RT-PCR method all of the samples in the special motif were positive. The deduced amino acid sequence of the cleavage site of the fusion (F) protein confirmed that all isolates contained the avirulent motif 112 GRQGRL117 at the cleavage site. All isolates have high similarity with LaSota and B1 strain in gene bank. The result in this study showed that migratory birds particularly aquatic birds in Bushehr carries lentogenic NDVs and act as one the reservoirs for the virus. So ND is always a great risk for the native avian population living on the routes of migratory birds.}, keywords = {Newcastle virus,Migratory birds,reverse transcription polymerase chain reaction (RT-PCR)}, title_fa = {بررسی مولکولی و پاتوتیپینگ ویروس های نیوکاسل جداشده از پرندگان مهاجر استان بوشهر}, abstract_fa = {پرندگان مهاجر و آبزی مخازن پارامیکسوویروس ها می باشند و عامل ایجاد کننده بیماری در مرغداری های صنعتی نیز محسوب میگردند، لذا بررسی مولکولی و پاتوتایپینگ ویروس های جدا شده از پرندگان مهاجر به منظور پیشگیری از انتقال این ویروس ضروری میباشد. نمونه های پژوهش حاضر از پرندگان مهاجر تالاب های استان بوشهر در جنوب کشور ایران جمع آوری شده است. تشخیص نمونه های جمع آوری شده و جداسازی ویروس در مؤسسه تحقیقات واکسن و سرم سازی رازی شیراز انجام شد. جهت بررسی مولکولی و شاخص های بیماری زایی 5 جدایه ویروس نیوکاسل، آزمایش های میانگین مدت زمان مرگ جنین جوجه (MDT)، شاخص بیماری زایی داخل مغزی (ICPI) و واکنش زنجیرهای پلی مراز معکوس (RT_PCR)انجام گردید. قطعه ی bp362 منطقه شکست پروتیین F با PCR تکثیر داده شد. تعیین توالی محصول PCR، جهت شناسایی اسیدهای آمینه و نوکلیوتیدها انجام شد. آنالیز فیلوژنی ناحیه شکست پروتئین F توسط برنامه های,Blast Bioeditو MegAlign-5 انجام گردید. بر اساس آزمایش های MDT، ICPI ،5 نمونه بررسی شده از سویه لنتوژن با حدت بیماری ضعیف تشخیص داده شد و همچنین در روش RT-PCR تمامی نمونه ها مورد مطالعه در منطقه اختصاصی bp 362 مثبت بودند. ردیف اسید های آمینه در منطقه شکست پروتیین F بصورت 112 GRQGRL117 می باشدکه همه لنتوژن می باشند. مقایسه انجام شده با تعدادی از ویروسها بانک ژنی، نشان داد بیشترین شباهت با ویروسهای لاسوتا و B1 می باشد. این تحقیق نشان داد که پرندگان مهاجر مخصوصا پرندگان آبزی در منطقه بوشهر می توانند یکی از ناقلین این ویروس باشند، بنابراین بیماری نیوکاسل همیشه یک خطر برای پرندگان بومی منطقه در مسیر پرندگان مهاجر می باشد.}, keywords_fa = {ویروس نیوکاسل,پرندگان مهاجر,واکنش زنجیرهای پلی مراز معکوس}, url = {https://www.ivj.ir/article_60932.html}, eprint = {https://www.ivj.ir/article_60932_69c2bd849e2f42bbf0cb0c6c1d5b47a2.pdf} }
