In recent years, this protein has been investigated to produce a recombinant vaccine. The aim of the present study was to construct a eukaryotic plasmid, expressing G1 epitope of BEFV G glycoprotein gene, for application as a possible DNA vaccine in future studies. For this purpose, the G1 epitope of G glycoprotein gene was cloned in a eukaryotic expression vector, pEGFP-N1, under the control of the human cytomegalovirus (CMV) promoter. Then, the recombinant pEGFPN1-G1 construct was transfected into human embryonic kidney (HEK 293) cell line and the expression efficiency was verified by indirect immunofluorescent (IFA) staining and immunoblotting techniques. Observation of intracytoplasmic fluorescence in transfected cells and appearance of a distinct band at an approximate molecular weight of 26 kDa in immunoblotting in reaction to an anti-G1 mouse serum, indicated that G1 protein was successfully expressed by this recombinant construct in the host cells.}, keywords = {Bovine ephemeral fever virus (BEFV),Recombinant G1 protein,DNA vaccine}, title_fa = {طراحی پلاسمید یوکاریوتی بیان کننده اپیتوپ G1 از ژن گلیکوپروتئین G ویروس تب بی ‏دوام گاوی در سلول ‏های جنینی کلیه انسان}, abstract_fa = {    گلیکوپروتئین G ویروس تب‏ بی‏دوام گاوی (BEFV) به عنوان یک نامزد احتمالی جهت ایمن‏سازی حیوانات در برابر بیماری تب بیدوام شناخته شده است.  در سال‏های اخیر، این پروتئین جهت تولید یک واکسن نوترکیب مورد توجه ویژه‏ بوده است.  هدف از مطالعهی حاضر، ساخت یک پلاسمید یوکاریوتی بیان کنندهی اپی‏توپ G1 از ژن گلیکوپروتئین G ویروس تب‏ بی‏دوام گاوی، برای استفاده به عنوان یک واکسن DNA احتمالی در مطالعات آینده بود.  به این منظور، اپی‏توپ G1 از ژن گلیکوپروتئین G در ناقل بیانی یوکاریوتی pEGFP-N1، تحت کنترل پروموتر سیتومگالوویروس انسانی (CMV) کلون شد.  سپس سازهی نوترکیب pEGFPN1-G1 به سلول‏های جنینی کلیهی انسان (HEK 293) انتقال یافت و کارایی بیان پروتئین با استفاده از روش‏های ایمونوفلورسنس غیرمستقیم (IFA) و ایمنوبلات بررسی شد.  مشاهدهی فلورسنس درون سیتوپلاسم سلول‏های ترانسفکت شده و ظهور یک باند مجزا با وزن مولکولی تقریبی 26 کیلو دالتون در آزمایش ایمنوبلات در واکنش به یک سرم موش ضد پروتئین G1، نشان‏دهندهی بیان موفق پروتئین G1 توسط این سازهی نوترکیب در سلول‏های میزبان بود.}, keywords_fa = {ویروس تب‏ بی‏دوام گاوی (BEFV),پروتئین نوترکیب G1,واکسن DNA}, url = {https://www.ivj.ir/article_57666.html}, eprint = {https://www.ivj.ir/article_57666_45d5377abf4fb530f8303687ac2d869a.pdf} }
