In this research, the effect of Lactobacillus rhamnosus (LGG) isolate with intestinal origin was studied on growth and enterotoxin production by Staphylococcus aureus and then, Staphylococcus aureus enterotoxin A gene expression was evaluated in a laboratory media. Therefore, this strain (1*107 cfu / ml) and Staphylococcus aureus (1*105  cfu / ml) were inoculated in TSB and  incubated in two temperatures at 25 and 35°C, at the time of zero, 24, 48 and 72 hours, the mixed cultures were counted in specific media. Staphylococcus aureus without lactobacillus was incubated as control and then enterotoxin production evaluated by using ELISA kit and the gene expression of the SEA of S. aureus in mixed and control cultures was also evaluated by RT Real Time PCR technique. The lactobacillus isolates decreased Staphylococcus aureus growth compared to control samples 3 log at 25°C and 2 log at 35°C. The enterotoxin production of A, C and E at 25° and enterotoxin type E and A at 35°C was inhibited by Lactobacillus rhamnosus. Comparison of gene expression SEA between treated and control cultures showed the gene expression of the SEA was significantly suppressed 6.82 fold in S. aureus co cultured with studied lactobacillus isolate at 25°C  while it was not significant to 35 °C (P<0.05). Because of the reduced effects of the isolates seen in this study, proposed it for natural prevention of bacterial growth/toxin production in foods to improve the quality and safety of the foods.}, keywords = {L. rhamnosus GG,enterotoxin A,Gene expression,S. aureus}, title_fa = {بررسی اثر مهاری لاکتوباسیلوس رامنوسوس سویه23527 LGG بر روی رشد و بیان ژنی انتروتوکسین A باکتری استافیلوکوکوس اورئوس ATCC 29213}, abstract_fa = {    افزایش ماندگاری، ایمنی مواد غذایی و ارتقای سلامتی انسان با استفاده از میکرو فلور طبیعی از موضوعات غالب در بهداشت عمومی محسوب می­شوند.  در این تحقیق اثر سویه­ی لاکتوباسیلوس رامنوسوس (LGG) با منشاء روده­ای بر رشد و تولید انتروتوکسین­های باکتری استافیلوکوکوس اورئوس و سپس بر بیان ژنی انتروتوکسین A باکتری استافیلوکوکوس اورئوس، در محیط آزمایشگاهی مورد بررسی قرار گرفت.  بدین منظور، پس از تلقیح این سویه (cfu/ml 107 × 1) و استافیلوکوکوس اورئوس (cfu/ml 105 × 1) در محیط TSB و گرم­خانه­گذاری در دو دمای 25 و °C 35، در زمان­های صفر، 24، 48 و 72 ساعت، از کشت مخلوط موجود در محیط TSB در محیط­های اختصاصی کشت و شمارش انجام شد.  استافیلوکوکوس اورئوس بدون حضور لاکتوباسیلوس به عنوان کنترل گرم­خانه­گذاری شد و سپس بررسی تولید انتروتوکسین با استفاده از کیت الیزای ریدااسکرین و همچنین بررسی بیان ژن انتروتوکسین A استافیلوکوکوس اورئوس در کشت­های مخلوط و کنترل به روش RT Real time PCR مطالعه گردید.  لاکتوباسیل مورد مطالعه، رشد استافیلوکوکوس اورئوس را نسبت به نمونه­ی کنترل 3 لوگ در دمای °C 25 و 2 لوگ در دمای °C 35 کاهش داد (05/0P<).  همچنین تولید انتروتوکسین­های A، C و E در دمای °C 25 و انتروتوکسین نوع E و A در دمای °C 35 توسط لاکتوباسیلوس رامنوسوس مهار گردید.  مقایسه­ی میزان بیان ژن sea بین کشت­های تیمار و کنترل در دمای °C 25 کاهش معنی­داری به میزان 82/6 برابر در بیان ژن نشان داد (05/0P<).  در حالی که کاهش معنی­داری در دمای °C 35 مشاهده نشد.  با توجه به اثر کاهشی مشاهده شده در این مطالعه، استفاده از این سویه برای پیش­گیری از رشد و تولید توکسین باکتریایی و همچنین جهت بهبود کیفیت و ارتقای سلامتی مواد غذایی پیشنهاد می­گردد.}, keywords_fa = {لاکتوباسیلوس رامنوسوس,استافیلوکوکوس اورئوس,انتروتوکسینA}, url = {https://www.ivj.ir/article_57665.html}, eprint = {https://www.ivj.ir/article_57665_955113daa4832f6eae05fd2bcf31a7b8.pdf} }
